Research Summary: My lab focuses on understanding the biochemical/molecular mechanisms underlying reduced immune function in aging. We primarily focus on identifying signaling pathways which may lead to changes in lipid metabolism in T-lymphocytes. We are currently using two mouse knockout models. One is a cbl-b knockout mouse to examine the cbl-mediated protein ubiquitination pathway and protein kinase C signaling in young and old rodent models. We are also using this model to assess the role of the T-lymphocyte in colon and pancreatic cancer development. The second model is the GPAT knockout mouse in which phospholipid biosynthesis is reduced. We use this model to understand the role of increased cholesterol to phospholipid ratios in altered membrane lipid raft and lipid signalling in thymic development and peripheral T-lymphocyte function. Our main goal is to determine how these signaling pathways lead to age-dependent alterations in phospholipid metabolism. Ultimately, we hope to develop dietary and/or pharmacological strategies to delay or offset reduced immune function in the elderly. Publications:
