Leica SP2 AOBS Confocal Microscope
Instrument:
Leica SP2 AOBS

The Leica SP2 AOBS confocal microscope can be used for fluorescence imaging of a variety of biological and materials samples. This instrument provides high resolution images using a variable pinhole for optical "z" sectioning and laser scanning.
Available objectives are shown below, with additional (high resolution) magnification is available with through digital zoom. The filterless scan head allow imaging of most common fluorescent probes. Excitation wavelengths available are 350 nm, 365nm, 458 nm, 476 nm, 488 nm, 496 nm, 514nm, 543 nm, 594 nm, and 633 nm.
Additional advanced techniques including spectral imaging, FRET, and FRAP are available with this instrument.
Objectives:
10X dry (HC PL APO 0.40NA CS)
20X dry (HC PL APO 0.70NA CS)
40X oil (HCX PL APO 1.25-.75NA CS)
63X oil (HCX PL APO 1.4-.60 NA Blau CS)
63X water (HCX PL APO 1.2NA Corr/0.17 CS)
The microscopy facility provides assisted use and training for this instrument.
For assisted use, contact a member of the microscopy staff to schedule an initial consultation. We will work with you to develop and experiment that is both technically feasible and meets your scientific needs.
For information about training, stop by the Microscopy Facility Office (1.426 H) and pick up a training request form and sign up for training seminars. Each trainee must first attend two seminars (no charge). These seminars discuss the theory of confocal microscopy and how to obtain high quality images. After you attend these seminars you may then schedule a “hands-on” training with either Dr. Bardo or John Mendenhall. The “hands-on” training will consist of at least 2-2hour session with training samples supplied by the Microscopy and Imaging Facility. Included in the training cost is 1-2 hour training session with your samples. For the 6 hours of “hands-on” training there is a flat fee of $100. Once training is complete the trained user will be given unassisted access to the instrument.
